QUANTIFICATION OF DNA DOUBLE-STRAND BREAKS IN BENIGN AND MALIGNANT BREAST DISEASES

Relevance: Double-strand DNA breaks are the most dangerous damage. Analysis of foci phosphorylated histone protein H2AX (γH2AX) is currently sensitive method for detecting double-strand breaks. This modification can become a biomarker cellular stress, especially in diagnosing and monitoring neoplastic diseases. In this study, we utilized novel pattern recognition algorithms on AKLIDES® platform to automatically analyze immunofluorescent images γH2AX compare results with visual scores. The formation peripheral blood mononuclear cells women breast cancer or benign tumors was studied.

 Results: When comparing indicators main control groups channel ruptures “FITC,” statistically significant difference found “Foci dia” (p=0.0382), “Focilnt mean” (p=0.0166), “Colocalisation” (p=0.0486). repair “AРС,” differences “Nuclei BGInt” (p=0.0166) indicator (p=0.0118).
 Conclusion: revealed changes along FITC break channels APC between possibly serve as diagnostic marker.
